Zinc is an essential trace mineral required as a catalytic cofactor for over 300 enzymatic reactions in the human body — more than any other trace mineral. It plays roles in virtually every physiological system.*

Zinc Orotate — enhanced cellular delivery — Zinc Orotate is zinc chelated with orotic acid (Vitamin B13) — the same orotic acid transport mechanism used in Priority One's Magnesium Orotate, Calcium Orotate, and Potassium Orotate products. Orotic acid is a naturally occurring compound involved in pyrimidine synthesis that serves as a molecular carrier for mineral transport across cell membranes.*

Orotic acid salts are theorised to improve cellular delivery of mineral ions by facilitating transport across the lipid bilayer — potentially providing superior intracellular zinc bioavailability compared to inorganic zinc salts (zinc oxide, zinc sulphate) and comparable or superior bioavailability to standard organic chelates (zinc citrate, zinc gluconate). The addition of zinc oxide in this blended form (zinc orotate oxide) provides additional elemental zinc concentration per capsule.*

Zinc at 340% DV — why a high-potency zinc supplement? — The DV for zinc (11mg for men, 8mg for women) represents the minimum requirement to prevent deficiency, not an optimal therapeutic or supplemental dose. Published research on zinc's clinical applications — including immune function, wound healing, hormone synthesis, and metabolic function — has typically used doses in the 25–50mg elemental zinc range. At 51mg (340% DV), Zinc Orotate provides a high-potency zinc dose appropriate for individuals with documented zinc insufficiency or those whose healthcare provider has recommended higher-dose zinc supplementation.*

Zinc and normal immune function — Zinc is one of the most important nutritional regulators of immune function. It is a required cofactor for the thymulin thymic peptide (which requires zinc for its biological activity), supports normal T-cell development and lymphocyte proliferation, normal natural killer cell activity, normal neutrophil function, and is directly required for over 100 immune-relevant zinc-finger transcription factors. Zinc deficiency — even marginal deficiency — impairs immune function significantly.*

Zinc and normal hormonal function — Zinc is specifically concentrated in the testes and prostate (the highest zinc concentrations in the male body), where it is required for normal testosterone synthesis, normal sperm production, and normal prostate cell metabolism. Zinc is also required for normal insulin synthesis and packaging in pancreatic beta cells (insulin is stored as a zinc-insulin hexamer), and supports normal growth hormone and IGF-1 signalling.*

Zinc and normal antioxidant defence — Zinc is a structural component of Cu/Zn-Superoxide Dismutase (Cu/Zn-SOD) — the primary cytoplasmic antioxidant enzyme that converts superoxide radical to hydrogen peroxide. Normal Cu/Zn-SOD activity depends directly on adequate zinc availability.*

Zinc and normal wound healing and tissue repair — Zinc is required for normal collagen synthesis (via prolyl hydroxylase enzyme activity), normal fibroblast proliferation, normal epithelial cell regeneration, and normal keratinocyte differentiation. It is one of the most important trace minerals for normal skin integrity and wound healing.*

Zinc and normal DNA synthesis and cell division — Zinc finger proteins are the largest family of transcription factors in the human genome — with over 700 zinc finger proteins regulating gene expression in virtually every cell type. 51 mg of zinc orotate does NOT equal 51 mg of elemental zinc

The negative reviews are ignorant to the facts that all zinc supplements aren't just 100% elemental zinc and have varying percentages of actual zinc because they're all made to be absorbed by our bodies in a way that elemental pure zinc cannot. Zinc orotate is actually around 15% actual zinc that can be absorbed by our bodies. The manufacturer should probably elaborate or make clear exactly what the percentage is.

Nowhere in the listing for this Zinc Orotate does it indicate the amount of zinc included in each capsule. I bought it anyway, thinking it the amount would be something reasonable/normal. Well, each capsule has 51mg of zinc. The upper-limit of zinc per day for adults is 40mg! Anything beyond that is too much, can be quite harmful. According to the National Institutes of Health, signs of too much zinc include "nausea, vomiting, loss of appetite, stomach cramps, diarrhea, and headaches. When people take too much zinc for a long time, they sometimes have problems such as low copper levels, lower immunity, and low levels of HDL cholesterol." I would therefore NOT recommend taking this Priority One Vitamins Zinc Orotate. I began experiencing low immunity, headaches, and stomach cramps after taking this, until I looked at the label and realized I was 25% above the daily limit for zinc.
