Folate as Optifolin+® (6S)-5-Methyltetrahydrofolate Choline Salt (850mcg DFE / 500mcg Folate, 213% DV)
Optifolin+® is a patented, registered trademark form of active folate from Balchem Corporation, delivering L-methylfolate as the choline salt — the biologically active form that enters the methylation cycle directly without requiring MTHFR enzyme conversion. Methylation is one of the three phases of oestrogen detoxification: after Phase I and Phase II liver metabolism convert oestrogens to water-soluble conjugates, methylation (catalysed by COMT — catechol-O-methyltransferase) converts reactive catechol oestrogen metabolites to their methylated, inactive forms for safe elimination. Without adequate methyl folate, this methylation step is impaired and potentially reactive oestrogen metabolites can accumulate.*

Calcium from Calcium D-Glucarate (50mg, 4% DV)
The calcium in this formula is delivered as calcium d-glucarate — the calcium salt of d-glucaric acid. It is the d-glucarate ion that is the pharmacologically active component, not the calcium. D-glucarate is a naturally occurring compound found in fruits and vegetables that supports Phase II liver detoxification through the glucuronidation pathway — the primary conjugation pathway through which oestrogens, environmental oestrogens, and other steroid hormones are rendered water-soluble and prepared for elimination.*
Specifically, d-glucarate inhibits beta-glucuronidase — a bacterial enzyme in the gut that deconjugates glucuronidated hormones, allowing them to be reabsorbed rather than eliminated. By inhibiting beta-glucuronidase, calcium d-glucarate supports the normal, complete elimination of conjugated oestrogens and other steroid hormones through the gut, reducing enterohepatic recirculation of oestrogens.*
Diindolylmethane (DIM, 50mg)
DIM is a compound formed in the stomach from indole-3-carbinol (I3C) — a compound found naturally in cruciferous vegetables (broccoli, Brussels sprouts, cauliflower, cabbage). DIM is the most bioavailable and active form of the indole class for hormonal support. DIM modulates Phase I cytochrome P450 enzymes (particularly CYP1A2) in the liver — the enzymes responsible for the initial hydroxylation of oestrogens. Specifically, DIM promotes 2-hydroxylation of oestradiol (producing 2-hydroxyoestrone — a more protective oestrogen metabolite) over 16-hydroxylation (which produces 16-alpha-hydroxyoestrone — a more oestrogenically active and less desirable metabolite).*
